چکیده

Purpose Transforming growth factor β (TGFβ) promotes cell survival by endorsing DNA damage repair and mediates an immunosuppressive tumor microenvironment. Thus, TGFβ activation in response to radiation therapy is potentially targetable because it opposes therapeutic control. Strategies assess this potential the clinic are needed. Methods Materials We evaluated positron emission tomography (PET) image 89Zr -fresolimumab, a humanized neutralizing monoclonal antibody, as means detect intracranial models. Pathway activity of was validated immunodetection phosphorylated SMAD2 target, tenascin. The contribution assessed Kaplan-Meier analysis mice bearing murine models GL261 SB28 glioblastoma brain-adapted 4T1 breast cancer (4T1-BrA) treated with 1D11, and/or focal (10 Gy). Results 89Zr-fresolimumab PET imaging detected engineered, physiological, radiation-induced activation, which confirmed immunostaining biological markers. tumors had greater signal compared similar-sized tumors, whereas widespread 4T1-BrA consistent their highly dispersed histologic distribution. Survival 1D11 antibody alone similar that control improved when given combination radiation. extent benefit associated degree PET. Conclusions This study demonstrates detects tumors. Autocrine growth inhibition by transforming growth factor β-1 (TGFβ-1) in human neuroendocrine tumour cells

Background and aim: The role of transforming growth factor β-1 (TGFβ-1) in neuroendocrine tumour biology is currently unknown. We therefore examined the expression and biological significance of TGFβ signalling components in neuroendocrine tumours (NETs) of the gastroenteropancreatic (GEP) tract. Methods: Expression of TGFβ-1 and its receptors, Smads and Smad regulated proteins, was examined in...

TGFβ (transforming growth factor β) receptor type III directs clathrin-mediated endocytosis of TGFβ receptor types I and II

The TGFβ (transforming growth factor β) pathway is an essential cell signalling pathway that is implicated in both normal developmental processes, such as organogenesis, and pathological disorders, such as cancer and fibrosis. There are three prototypical TβRs (TGFβ receptors): TβRI (TβR type I), TGβRII (TβR type II) and TGFβRIII (TβR type III, also known as betaglycan). Whereas the role of TβR...

The effect of fasting on Positron Emission Tomography (PET) imaging

As a nuclear approach, Positron Emission Tomography (PET) is a functional imaging technique which is based on the detection of gamma ray pairs emitted by a positron-emitting radionuclide. There are certain limitations to this technique such as normal tissue uptake. Therefore, it has been recommended that patients prepare before scanning. Fasting for a short while before PET imaging is an exampl...

Imaging Cellular Proliferation in Prostate Cancer with Positron Emission Tomography

Prostate cancer remains a major public health problem worldwide. Imaging plays an important role in the assessment of disease at all its clinical phases, including staging, restaging after definitive therapy, evaluation of therapy response, and prognostication. Positron emission tomography with a number of biologically targeted radiotracers has been demonstrated to have potential diagnostic and...

Silencing of the Transforming Growth Factor-β (TGFβ) Receptor II by Krüppel-like Factor 14 Underscores the Importance of a Negative Feedback Mechanism in TGFβ Signaling*S⃞

The role of non-Smad proteins in the regulation of transforming growth factor-beta (TGFbeta) signaling is an emerging line of active investigation. Here, we characterize the role of KLF14, as a TGFbeta-inducible, non-Smad protein that silences the TGFbeta receptor II (TGFbetaRII) promoter. Together with endocytosis, transcriptional silencing is a critical mechanism for down-regulating TGFbeta r...
